Assessment of four different detergents used to extract membrane proteins from Xylella fastidiosa by two-dimensional electrophoresis BJM
Di Ciero,Luciana; Bellato,Cláudia de M.; Meinhardt,Lyndel W.; Ferrari,Fernanda; Castellari,Rafael R.; Marangoni,Sérgio; Novello,José C..
Four different detergents, ASB 14, SB 3-10, CHAPS and Triton X100, were utilized to determine the optimal detergent for the solubilization of membrane proteins from the phytopathogenic bacterium Xylella fastidiosa. These proteins were differentially solubilized in distinct buffers containing the detergent and subjected to bidimensional electrophoresis within the non-linear pH range of 3-10. The detergents ASB 14 and SB 3-10 were the most effective revealing 221 and 157 spots, respectively. CHAPS and Triton X100 were less effective and revealed only 72 and 43 spots, respectively. MALDI-TOF tryptic peptide mass fingerprinting of 18 excised proteins from the ASB 14 treatment revealed that 83% were membrane proteins and that the theoretical efficiency of...

Deciphering the Infectious Process of Colletotrichum lupini in Lupin through Transcriptomic and Proteomic Analysis ArchiMer
Dubrulle, Guillaume; Picot, Adeline; Madec, Stephanie; Corre, Erwan; Pawtowski, Audrey; Baroncelli, Riccardo; Zivy, Michel; Balliau, Thierry; Le Floch, Gaétan; Pensec, Flora.
The fungal phytopathogen Colletotrichum lupini is responsible for lupin anthracnose, resulting in significant yield losses worldwide. The molecular mechanisms underlying this infectious process are yet to be elucidated. This study proposes to evaluate C. lupini gene expression and protein synthesis during lupin infection, using, respectively, an RNAseq-based transcriptomic approach and a mass spectrometry-based proteomic approach. Patterns of differentially-expressed genes in planta were evaluated from 24 to 84 hours post-inoculation, and compared to in vitro cultures. A total of 897 differentially-expressed genes were identified from C. lupini during interaction with white lupin, of which 520 genes were predicted to have a putative function, including...

Different secretory repertoires control the biomineralization processes of prism and nacre deposition of the pearl oyster shell ArchiMer
Marie, Benjamin; Joubert, Caroline; Tayale, Alexandre; Zanella-cleon, Isabelle; Belliard, Corinne; Piquemal, David; Cochennec-laureau, Nathalie; Marin, Frederic; Gueguen, Yannick; Montagnani, Caroline.
Mollusca evolutionary success can be attributed partly to their efficiency to sustain and protect their soft body with an external biomineralized structure, the shell. Current knowledge of the protein set responsible for the formation of the shell microstructural polymorphism and unique properties remains largely patchy. In Pinctada margaritifera and Pinctada maxima, we identified 80 shell matrix proteins, among which 66 are entirely unique. This is the only description of the whole "biomineralization toolkit" of the matrices that, at least in part, is thought to regulate the formation of the prismatic and nacreous shell layers in the pearl oysters. We unambiguously demonstrate that prisms and nacre are assembled from very different protein repertoires....

Egg quality in domesticated and wild seabass (Dicentrarchus labrax): A proteomic analysis ArchiMer
Crespel, Amlie; Rime, Helene; Fraboulet, Erwann; Bobe, Julien; Fauvel, Christian.
In order to improve Our understanding of factors determining egg quality in fish, seabass egg batches obtained from 34 females held under standard conditions, were assessed for biological performances and a fraction from each batch was subjected to proteomic analysis. Comparison of proteomic patterns from eggs of different quality revealed differential protein spots among which one was identified as Nucleoside Diphosphate Kinase, a highly conserved protein involved in cell proliferation. This study shows that molecular markers may help understand factors determining egg quality in fish.

High pressure-sensitive gene expression in Lactobacillus sanfranciscensis BJMBR
Vogel,R.F.; Pavlovic,M.; Hörmann,S.; Ehrmann,M.A..
Lactobacillus sanfranciscensis is a Gram-positive lactic acid bacterium used in food biotechnology. It is necessary to investigate many aspects of a model organism to elucidate mechanisms of stress response, to facilitate preparation, application and performance in food fermentation, to understand mechanisms of inactivation, and to identify novel tools for high pressure biotechnology. To investigate the mechanisms of the complex bacterial response to high pressure we have analyzed changes in the proteome and transcriptome by 2-D electrophoresis, and by microarrays and real time PCR, respectively. More than 16 proteins were found to be differentially expressed upon high pressure stress and were compared to those sensitive to other stresses. Except for one...

Is pallial mucus involved in Ostrea edulis defenses against the parasite Bonamia ostreae? ArchiMer
Fernández-boo, Sergio; Gervais, Ophelie; Prado-alvarez, Maria; Chollet, Bruno; Claverol, Stéphane; Lecadet, Cyrielle; Dubreuil, Christine; Arzul, Isabelle.
Bonamia ostreae is an intrahemocytic parasite that has been responsible for severe mortalities in the flat oyster Ostrea edulis since the 1970́s. The Pacific oyster Crassostrea gigas is considered to be resistant to the disease and appears to have mechanisms to avoid infection. Most studies carried out on the invertebrate immune system focus on the role of hemolymph, although mucus, which covers the body surface of molluscs, could also act as a barrier against pathogens. In this study, the in vitro effect of mucus from the oyster species Ostrea edulis and C. gigas on B. ostreae was investigated using flow cytometry. Results showed an increase in esterase activities and mortality rate of parasites exposed to mucus from both oyster species. In order to...

Proteome of fraction from Tityus serrulatus venom reveals new enzymes and toxins J. Venom. Anim. Toxins incl. Trop. Dis.
Amorim,Fernanda Gobbi; Longhim,Heloisa Tavoni; Cologna,Camila Takeno; Degueldre,Michel; Pauw,Edwin De; Quinton,Loïc; Arantes,Eliane Candiani.
Abstract Background: Tityus serrulatus venom (Ts venom) is a complex mixture of several compounds with biotechnological and therapeutical potentials, which highlights the importance of the identification and characterization of these components. Although a considerable number of studies have been dedicated to the characterization of this complex cocktail, there is still a limitation of knowledge concerning its venom composition. Most of Ts venom studies aim to isolate and characterize their neurotoxins, which are small, basic proteins and are eluted with high buffer concentrations on cation exchange chromatography. The first and largest fraction from carboxymethyl cellulose-52 (CMC-52) chromatography of Ts venom, named fraction I (Fr I), is a mixture of...

Ben Amor, Faten; Elleuch, Fatma; Ben Hlima, Hajer; Garnier, Matthieu; Saint-jean, Bruno; Barkallah, Mohamed; Pichon, Chantal; Abdelkafi, Slim; Fendri, Imen.
The green microalgae Dunaliella genus is known for the production of high added value molecules. In this study, strain AL-1 was isolated from the Sebkha of Sidi El Hani (Sousse, Tunisia). This isolate was identified both morphologically and genetically via 18S rRNA gene sequence as a member of the genus Dunaliella. Strain AL-1 was found to be closely related to Dunaliella salina, Dunaliella quartolecta and Dunaliella polymorpha with more than 97% similarity. Response surface methodology was used to maximize carotenoid production by strain AL-1 by optimizing its growth conditions. The highest carotenoid content was obtained at salinity: 51, light intensity: 189.89 μmol photons·m−2·s−1, and nitrogen: 60 mg·L−1. Proteomic profiling, using two-dimensional gel...

The inhibitory effect of calcium on Cylindrospermopsis raciborskii (cyanobacteria) metabolism BJM
Carneiro,Ronaldo Leal; Alípio,Ana Carla Nascimento; Bisch,Paulo Mascarello; Azevedo,Sandra Maria Feliciano de Oliveira e; Pacheco,Ana Beatriz Furlanetto.
Cylindrospermopsis raciborskii (Woloszynska) Seenaya & Subba Raju is a freshwater cyanobacterium of worldwide distribution. In the North-eastern region of Brazil many eutrophic water reservoirs are characterized by the dominance of C. raciborskii, with recurrent occurrence of blooms. These water bodies have high conductivity due to a high ionic concentration, and are defined as hard (with high values of CaCO3). In this study, we investigated the long-term effect (12 days) of high calcium concentration (8 mM Ca2+) on C. raciborskii (T3 strain) growth, morphology, toxin content, and metabolism. Changes in protein expression profiles were investigated by proteomic analysis using 2D gel electrophoresis and mass spectrometry. A continued exposure to...

Trypanosoma cruzi: analysis of two different strains after piplartine treatment BJID
Vieira,Gabriela Alves Licursi; Silva,Marco Túlio Alves da; Regasini,Luis Octávio; Cotinguiba,Fernando; Laure,Helen Julie; Rosa,José César; Furlan,Maysa; Cicarelli,Regina Maria Barretto.
ABSTRACT The hemoflagellate protozoan, Trypanosoma cruzi, mainly transmitted by triatomine insects through blood transfusion or from mother-to-child, causes Chagas' disease. This is a serious parasitic disease that occurs in Latin America, with considerable social and economic impact. Nifurtimox and benznidazole, drugs indicated for treating infected persons, are effective in the acute phase, but poorly effective during the chronic phase. Therefore, it is extremely urgent to find innovative chemotherapeutic agents and/or effective vaccines. Since piplartine has several biological activities, including trypanocidal activity, the present study aimed to evaluate it on two T. cruzi strains proteome. Considerable changes in the expression of some important...
